Title: Evaluation of hydrogels of methacrylated inulin and methacrylated azo inulin as excipients for colonic drug targeting

Abstract: Different types of hydrogels consisting of either methacrylated inulin (MA-IN) or a mixture of methacrylated inulin, 2-hydroxyethylmethacrylate or methacrylic acid and additional azo a romatic cross-links were synthesized, and their biodegradable properties were evaluated by determination of the equilibrium degree of swelling and the amount of released fructose, after incubation in a system based on rat or human faecal bacteria. MA-IN hydrogels prepared from 27% w/w solutions of MA-IN and a degree of substitution of 8.1, or prepared from 22% w/w solutions of MA-IN and a degree of substitution of 12.1 are susceptible to bulk degradation both in a rate caecal and a human faecal degradation medium. Apparently, hydrogels attain a maximum extent of degradation, depending on their feed composition, within an incubation period of 24 h, regardless of whether the degrading enzymes are located intracellularly or not. Hydrogels containing azo aromatic cross-links were not degraded due to increased hydrophobicity and increased network density.
