Abstract: In the immunohistochemical analysis of the brain, tissue preparation and fixation are critical steps. During our studies of transplanting human bone marrow multipotent adult progenitor cells (hMAPCs) into the rat brain, we noticed that various methods of brain tissue preparation and fixation differentially influenced antigenic preservation in the transplants and in the host brain. Here, we report a simple, effective and reproducible method of tissue preparation and fixation that results in the immunohistochemical labeling of transplanted and host cells.
