Abstract: The study of conformationally restricted carbohydrate modified nucleic acids has given new insights into the concept of the antisense technology. We learned to understand the structural requirements of a modified nucleic acid to function as steric blocker for RNA. Several of the physicochemical and conformational factors influencing duplex stabilization are analyzed with respect to their relative importance for the antisense field.
